Proteoform Thursday: Byron Drown presents “Approaches in Targeted Top-Down Proteomics”

Characterization of proteoforms from complex mixtures by LC-MS/MS is a powerful method for observing dynamic modifications. Even with advances in separation efficiency, MS sensitivity and speed, and data analytics, however, many lower-abundance proteoforms remain just below the limit of detection. Immunoprecipitation can provide a high degree of enrichment in a single step, yet each antibody-antigen pair often requires significant optimization. We established a platform for rapidly developing targeted top-down proteomics assays with transportable sample preparation methods. With more robust sample preparation methods and a shorter time between experiment design and proteoform identification, the scope of proteoforms accessible by top-down mass spectrometry has greatly expanded.
