Proteoform Thursday: Fanny Caroline Liu presents “Advancing Top-Down Protein Analysis by Tandem-TIMS/MS Approaches”

We recently developed a novel tandem-trapped ion mobility spectrometry/mass spectrometry (tandem-TIMS/MS) instrument capable of performing collision-induced dissociation (CID) and ultraviolet photodissociation (UVPD). Here, we (1) utilize tandem-TIMS-CID to investigate subunits of native, intact protein complexes, (2) conduct top-down protein sequencing by integration of parallel accumulation serial fragmentation (PASEF) with tandem-TIMS-UVPD measurements, and (3) investigate the cross sections of top-down protein fragment ions by time-resolved measurements in tandem-TIMS/MS.
